OBP3 Expression Levels and Adult Phenotypes of OBP3::OBP3-GUS Transgenic Lines. Arabidopsis was transformed with a construct containing genomic DNA encoding OBP3 translationally fused to the GUS gene. Expression of this fusion gene was controlled by approximately 1.4kb of DNA 5` of the OBP3 gene. (A) Total RNA was isolated from five-day-old seedlings grown in continuous white light. PCR was performed on the cDNA using OBP3 and GUS specific primers for the lines containing the OBP3-GUS gene, and OBP3 specific primers for the control lines. PCR using OBP3 and OBP3-GUS primers was amplified for 32 cycles. UBQ1 cDNA amplified for 24 cycles was used to normalize the cDNA template. (B) Four-week-old, homozygous lines transformed with the OBP3::OBP3-GUS fusion protein were grown in long-day conditions. All lines have a single locus insertion.
