• Supplemental Figure 1 - Supplemental Figure 1. Single cells expressing GFP on the surface of biolistically transformed callus. (A) Non-transformed cells without GFP expression; (B) Transformed cells with GFP expression. The area visible here is equivalent to ~5% of each panel of Figure 1C.
• Supplemental Figure 2 - Supplemental Figure 2. Bisulfite genomic DNA methylation sequencing. See Figure 2 legend: a second, independent, set of sequencing reactions was undertaken. Eleven clones are shown for IR24 (A) and 15 clones for T309 (B)
• Supplemental Figure 3 - Supplemental Figure 3. Genomic DNA blot analysis of T309 and IR24. Genomic DNA (2 μg) was digested with HindIII and separated on a 1% agarose gel before transfer to a nylon membrane. The blot was probed with the rubq2 3' UTR sequence. The upper band in both the T309 and IR24 lanes is a rubq2 genomic DNA fragment. The white arrowhead indicates the presence of an additional fragment of rubq2 3' UTR in T309. A DNA size marker is shown on the left.
