Supplemental Figure 1 -
Supplemental Figure 1. Kinetics for expression of the integrated DR5:GUS reporter gene in response to NAA and IAA in wild type Arabidopsis mesophyll protoplasts. Protoplasts were treated with 1μM 1-NAA (left panels) or 1μM IAA (right panels) for the times indicated.
(A) Quantitative GUS activities were measured at the times indicated following auxin treatment. Standard errors are indicated. In some cases, error bars are not visible because of the small size of the error bars.
(B) RT-PCR was performed at the times indicated following auxin treatment. Expression of the natural ACT7 gene was used as a standard.
(C) Real time RT-PCR was performed at the times indicated following auxin treatment. Expression of the natural ACT7 gene was used as a standard.

(A) Protoplasts were isolated from wild type, arf6-2, nph4-1, arf8-2 and arf19-3 mutant leaves, transfected with the DR5:GUS reporter gene and treated with 1μM 1-NAA for 20-22 h before GUS activities were measured. Standard errors are indicated. In some cases, error bars are not visible because of the small size of the error bars.
(B) Same as (A), but protoplasts were transfected with 2XD0:GUS reporter gene.

Supplemental Figure 3. Expression of the IAA1 and IAA19 natural auxin response genes in nph4-1 Arabidopsis mesophyll protoplasts transfected with effector genes encoding full-length ARF activators.
Protoplasts were isolated from nph4-1 leaves, transfected with 35S:ARF effector genes or 35S:CAT, and treated (black columns) or not treated (white columns) with 1μM 1-NAA for 20-22h. Total RNA was isolated, and real time RT-PCR was used to test the expression levels of the natural auxin response genes. Expression of the natural ACT7 gene was used as a control. Expression levels are presented as values relative to that of 35S:CAT transfected mock treated sample, defined as 1, after normalization to ACT7 levels.
