Supplemental Figure 1

Supplemental Figure 1. Sequence comparison of Arabidopsis AtETFQO with ETFQO homologues in various organisms: OsETFQO, Oryza sativa; HsETFQO, human homo sapiens; MmETFQO, mouse Mus musculus; DmETFQO, fly Drosophila melanogaster; CeETFQO, worm Caenorhabditis elegans; ScETFQO, yeast Saccaromyces cerevisiae; LpETFQO, proteobacteria Legionella pneumophila. The residues in black indicate amino acids that are identical to those of Arabidopsis ETFQO in the alignment, and those in gray indicate similar amino acids to those of Arabidopsis ETFQO. The red box near the N-terminus indicates a putative binding domain for the ADP moiety of flavin (Goodman et al., 1994), which was highly conserved among all the species. The blue box near the C-terminus indicates a C-X23-C-X2-C-X2-C sequence, which is commonly found in proteins with [4Fe-4S]2+1+ clusters, with the four cysteine residues postulated to act as ligands for the iron atoms of the cluster (Goodman et al., 1994). The red triangle near the N-terminus indicates a putative cleavage position of the mitochondrial target peptide predicted by the PSORT program (Nakai and Horton, 1999). The positions of the four cysteine residues that putatively ligand the [4Fe-4S]2+1+ cluster were also highly conserved among species. Alignment was performed using ClastalW (http://www.ebi.ac.uk/clustalw/). Boxshading was produced by BOXSHADE 3.21 (http://ch.embnet.org/software/BOX_form.html).

Supplemental Figure 2. Relative levels of the metabolites, succinate, fumarate, malate, and pyroglutamate, in wild type (black bars) and etfqo-1 mutant (white bars) during extended dark condition. Y-axis values represent metabolite level relative to wild type. Data were normalized to mean response calculated for the 0 day dark treated leaves of wild type. Values presented are mean ± SE of determinations on six individual plants.
