Pictures were taken 3 days after infiltration. (B) Nt CMPG1W64A protein levels were analyzed by western blot. • Supplemental Figure 2 - Tobacco CMPG1 Is Involved in Inf1-Mediated HR Production. (A) Leaf panels of Cf9 tobacco control and Cf9 tobacco overexpressing Nt CMPG1 or Nt CMPG1C37A were infiltrated with agrobacteria delivering Inf1 at the indicated cell densities. Pictures were taken 4 days after infiltration. (B) Cf9 tobacco control and two lines of Cf9 tobacco carrying Nt CMPG1 “hairpin” were infiltrated with the indicated dilutions of IF(+Avr9) for HR production. Pictures were taken 3 days after infiltration. (C) Cf9 tobacco control and two lines of Cf9 tobacco carrying Nt CMPG1 “hairpin” were infiltrated with the indicated optical density of agrobacteria carrying Inf1. Pictures were taken 4 days after infiltration. (D) Leaf disks from the indicated tobacco lines were harvested 30 minutes after elicitation with IF(+Avr9) and Nt CMPG1 levels were analyzed by RT-PCR. • Supplemental Figure 3 - Correlation Between Tomato Cmpg1 mRNA Levels and Defense Responses in Tomato “Hairpin” Lines. Cf9 tomato plants were transformed with pHellsgate containing a tomato Cmpg1 “hairpin” and 3 lines were selected for further analysis. (A) Cf9 untransformed and 3 transgenic lines were infiltrated with IF(+Avr9) for HR production. Pictures were taken 3 days after infiltration. (B) The same tomato lines as in (A) were infiltrated with IF(+Avr9) and leaf disks were harvested 30 minutes after infiltration for Cmpg1 mRNA amplification (left panel), or 24 hours after infiltration for PR5 mRNA amplification (right panel). Correlation was observed among HR production, Cmpg1 mRNA levels and PR5 mRNA levels. Complete C. fulvum infection was only observed in the line with the lowest Cmpg1 mRNA induction (line E).
