Supplemental Figure 1 -
Analysis of FIS2 imprinting at 3 DAP and 5 DAP. Allele-specific RT-PCR on RNAs extracted from siliques obtained at 3 or 5 DAP after crosses between wild-type Col and several mutants affected for DNA methylation. A 180 bp deletion in the FIS2 coding sequence of the wild-type accession Col allows distinction of the Col FIS2 transcript from the FIS2 transcript in accessions C24. The paternal (p) and maternal (m) alleles are indicated.
Supplemental Figure 2 -
Bisulfite sequencing of the FIS2 5’ region contains a 200 bp domain enriched in methylated cytosine residues predominantly on CpGs. Analysis was done with genomic DNA from rosette leaves. Analysis was done on wild-type and met1-3/met1-3 plants. Numbers are from the putative translation site. Each line of circles represents the result from bisulfite sequencing of a single clone. Filled circles represent methylated cytosine. Open circles, unmethylated cytosine.
